Phosphorolytic Cleavage of Fructose-&phosphate by Fructose-6-phosphate Phosphoketolase from Acetobacter xylinum*
نویسنده
چکیده
The synthesis of cellulose by washed suspensions of Acetobatter xylinum proceeds in the presence of iodoacetate at concentrations which completely inhibit the oxidation of glyceraldehyde-a-phosphate (1). Since glucose was the only carbon compound added, the possibility was considered that the energy required for the biosynthetic process may be derived from reactions linked to the pentose phosphate cycle which was shown to be operative in A. zylinum (2). An exploration of energyyielding processes in extracts of A. xylinum revealed the presence of an enzyme which catalyzes the reaction shown in Equation I.

متن کاملCrystal structures of phosphoketolase: thiamine diphosphate-dependent dehydration mechanism.
Thiamine diphosphate (ThDP)-dependent enzymes are ubiquitously present in all organisms and catalyze essential reactions in various metabolic pathways. ThDP-dependent phosphoketolase plays key roles in the central metabolism of heterofermentative bacteria and in the pentose catabolism of various microbes. In particular, bifidobacteria, representatives of beneficial commensal bacteria, have an e...
متن کاملRegulation of hexose phosphate metabolism in Acetobacter xylinum.
The metabolism of glucose and fructose was studied in resting succinate-grown cells of Acetobacter xylinum. From fructose only cellulose and CO(2) were formed by the cells, whereas from glucose, gluconate was formed much more rapidly than these two products. The molar ratio of sugar converted into cellulose to sugar converted into CO(2) was significantly greater than unity for both hexoses. The...

Cell-free extracts of 17 strains of Bifidobacterium bifidum (Lactobacillus bifidus) were examined for the presence of aldolase, glucose-6-phosphate dehydrogenase, and fructose-6-phosphate phosphoketolase. All strains turned out to lack aldolase, an enzyme unique to glycolysis, and glucose-6-phosphate dehydrogenase, characteristic of the hexosemonophosphate pathway. In all strains, fructose-6-ph...

Ryruvate-phosphate dikmase formation was induced on transfer of glucose-grown cells of Acefobacfer xylinum to succinateor pyruvate-containing media. Enzyme formation did not occur if such media were supplemented with glucose or in the presence of chloramphenicol. Fructose and glycerol were less effective in repressing enzyme formation. When glucose was added to cultures of cells growing on succ...
